Efficacy of Biologics Targeting Tumour Necrosis Factor-alpha, Interleukin-17 -12/23, -23 and Small Molecules Targeting JAK and PDE4 in the Treatment of Nail Psoriasis: A Network Meta-analysis

The introduction of biologics and small molecules has revolutionized the treatment of psoriasis. This paper aimed to determine the comparative efficacy of biologics and small molecules in nail psoriasis, a ‘difficult- to-treat’ condition. To synthesize evidence from already published data, we performed a network meta- analyses. Based on 17 randomized clinical trials, IL-17 inhibitor ixekizumab seems to be the best treatment option, whereas the IL-12/23 inhibitor ustekinumab and the IL-23 inhibitor guselkumab are less effective. We hope this paper can aid dermatologists in choosing the best therapeutic option for nail psoriasis and will inspire further research on the long-term efficacy of these treatments.
