We performed largescale gene-expression profiling in patients with mycosis fungoides (MF), and to our surprise we found that B-cells were significantly upregulated in the microenvironment in MF compared to controls. We could confirm the presence of B-cells in situ and this raised a number of important questions. How does the infiltrating B-cells impact MF biology, and do they have a role in disease progression? Can we re-establish immunological tumor control by targeting B-cells therapeutically? Further studies are required to elucidate the role of B-cells in MF and our group have initiated several investigations exploring the nature of B-cell infiltration in MF.

Clinicopathological, Genetic and Survival Advantages of Naevus-associated Melanomas: A Cohort Study
Xavier Bosch-Amate, Sebastian Podlipnik, Constanza Riquelme-Mc Loughlin, Cristina Carrera, Alicia Barreiro-Capurro, Adriana García-Herrera, Llucia Alós, Josep Malvehy, Susana Puig
Several studies have suggested that naevus-associated melanomas differ from de novo melanomas, being thinner and with less ulceration; however, the prognostic implication is unclear. The objective of this study was to describe clinicopathological, genetic and survival characteristics of de novo and naevus-associated melanomas in a cohort of primary invasive cutaneous melanomas over a 20-year perio ...

This study evaluated whether secukinumab treatment for patients with moderate to severe plaque psoriasis correlates with improvements in symptoms of anxiety and depression. SUPREME was a 24-week, phase IIIb, multicentre, prospective study conducted across 50 centres in Italy with an extension period of up to 72 weeks. Assessments used were: Psoriasis Area Severity Index (PASI), Hospital Anxiety ...

Incomplete excisions of melanocytic lesions occur despite the intention of complete removal. The aim of this study was to determine the incomplete excision rates for benign and malignant melanocytic lesions and the associated risk factors. Demographic, clinical, and histopathological data possibly associated with incomplete excision were collected from 2,782 consecutive excisions between 2014 an ...

Diagnosis of Epidermolysis Bullosa Acquisita: Multicentre Comparison of Different Assays for Serum Anti-type VII Collagen Reactivity

Epidermolysis bullosa acquisita is a pemphigoid disease characterized by autoantibodies against type VII collagen. This study compared the sensitivity and specificity of 6 diagnostic assays: type VII collagen non-collagenous domains enzyme-linked immunoassay (NC1/2 ELISA) (MBL, Nagoya, Japan); type VII collagen NC1 ELISA (Euroimmun, Lübeck, Germany); indirect immunofluorescence (IF) microscopy te ...

Immunomodulator Galectin-9 is Increased in Blood and Skin of Patients with Bullous Pemphigoid
Jasper Pruessmann, Wiebke Pruessmann, Maike M. Holtsche, Beke Linnemann, Christoph M. Hammers, Nina van Beek, Detlef Zillikens, Enno Schmidt, Christian D. Sadik
Massive recruitment of eosinophils into the dermis is a hallmark of bullous pemphigoid pathogenesis. Identifying the chemoattractant(s) guiding eosinophils into the skin in bullous pemphigoid is a prerequisite to therapeutic targeting of eosinophil recruitment. Galectin -9 is a potent chemoattractant for eosinophils, but its potential role in bullous pemphigoid is unknown. The aim of this study ...

Hypertrophic Scar Outcomes in Fractional Laser Monotherapy Versus Fractional Laser-Assisted Topical Corticosteroid Delivery: A Randomized Clinical Trial

Topical corticosteroid delivery following fractional laser treatment is an effective means of treating hypertrophic scars. However, the relative efficacy of adjuvant corticosteroid treatment vs fractional laser monotherapy alone is unclear. The aim of this study was to compare the efficacy and safety of fractional laser-assisted topical corticosteroid delivery with fractional laser monotherapy i ...

Role of B-cells in Mycosis Fungoides
Pia Rude Nielsen, Jens Ole Eriksen, Mia Dahl Sørensen, Ulrike Wehkamp, Lise M. Lindahl, Michael Bzorek, Lars Iversen, Anders Woetman, Niels Ødum, Thomas Litman, Lise Mette Rahbek Gjerdrum
Mycosis fungoides is the most common type of cutaneous T-cell lymphoma. The inflammatory microenvironment in mycosis fungoides is complex. There is accumulating evidence that the neoplastic T-cells take control of the microenvironment and thereby promote their own expansion by suppressing cellular immunity. B-cells have proved to be upregulated in large-cell transformed mycosis fungoides, and co ...

Diagnostic Value and Practicability of Serration Pattern Analysis by Direct Immunofluorescence Microscopy in Pemphigoid Diseases
Maike M. Holtsche, Nina van Beek, Axel Künstner, Hauke Busch, Detlef Zillikens, Enno Schmidt
In pemphigoid diseases, direct immunofluorescence can be used to differentiate 2 patterns of antibody deposition at the dermal–epidermal junction; u- and n-serrated pattern. The u-serrated pattern is found in epidermolysis bullosa acquisita, and n-serrated pattern in all other pemphigoid diseases. To determine the detection frequency of these serrated patterns and the optimal thickness of biopsy ...
